Integrity, Precautions and Filtration Pressure of Membrane Filter
Integrity, Precautions, and Filtration Pressure of Membrane Filter
Flow Filtration of Membrane Filter
In order to effectively intercept the target molecule and ensure the yield, it is generally recommended to select the 1/3-1/5 membrane pore size of the target sample molecule. At the same time, in order to minimize the contaminant protein content during the concentration and diafiltration process, the yield of the target molecule should be the first choice. The largest possible aperture is under guarantee conditions. If you want to use a tangential flow filtration membrane to separate two target molecules of different sizes, in addition to considering the size of the sample molecule, the choice of pore size will also be affected by the uniformity of the membrane pore size.
Product Yield of Membrane Filter
The design is reasonable, the filter part of the filter is made of glass, the chemical performance is stable, and the sealing degree is strong, which can effectively prevent external pollution and ensure the success rate of bacterial detection. Generally speaking, the more uniform the membrane pore size is, the filter membrane closer to the molecular weight of the target molecule can be selected for concentration, so that the permeation rate is faster, the product yield is higher, and the impurities are easier to wash out.
Commissioning Operation for Better Filtration Effect
The exterior of the host is scrubbed clean with alcohol. Effective grounding is necessary to protect and maintain the instrument. Select the filter membrane raw materials based on the properties of the test product, and ensure the integrity of the filter membrane before and after filtration. When the instrument is not working, disconnect the power supply. When the instrument is on standby, close the solenoid valve and set it to “off” status. Strong acid, strong base, a strong oxidant, strong corrosive, and other liquids cannot be filtered. When the test product contains insoluble particles and suspended matter, it may cause the filter membrane to block and affect the filtration. The test solution should be pretreated to remove the particles of suspended matter.
Practical Procedures in Filtration
The microfiltration membrane and the disposable microbial limit filters are connected with the filter head to ensure the integrity of the microporous filter membrane. Pour the test solution prepared in advance into the filter cup and do not manipulate the scale of the filter cup. Press the “power” button, then open the corresponding valve, and then press the “power” button to complete the collection of bacteria. After all the test solution is filtered, close the corresponding valve, and then press the “power” button to complete the collection.
Precautions of Membrane Filter
The filter membrane should be used in a microporous membrane filter with a ring gasket with good sealing performance and a membrane support plate with better rigidity. Since the film is sterilized by ethylene oxide, when the package is opened, there may still be a trace of alcohol-like odor, which has no effect on the quality of the film. Before filtering the liquid medicine, add an appropriate amount of water for injection to moisten and filter, and check the clarity of the filtrate and the integrity of the membrane.
Filtration Pressure of Membrane Filter
The filtration pressure is used in accordance with the regulations in the corresponding SOP and generally does not exceed 0.1Mpa. The membrane is flammable. It should not be near a fire during transportation and storage. It should not be touched by sharp objects. In order to prevent the filter membrane from being damp and deteriorated, it should be stored in the dark at room temperature and relative humidity of about 60%. The filter membrane loses water and curls, and it can still be used after soaking. Check the filter membranes one by one, and the filter membranes without pinholes, transparent spots and foreign objects can be used.
